SOCS3
SOCS3사이토카인 시그널링 억제제 3(SOCS3 또는 SOCS-3)은 SOCS3 [5][6]유전자에 의해 인체 내에서 암호화되는 단백질이다.이 유전자는 사이토카인 시그널링 억제제(SOCS)로도 알려진 STAT 유도 STAT 억제제(SSI)의 일원을 코드한다.SSI 패밀리는 사이토카인 유도형 음성조절제이다.
SOCS3는 보존된 유전자로, 드로소필라,[7][8] 닭, [9]악어를 포함한 동물의 왕국에서 발견된다.
기능.
SOCS3 유전자의 발현은 IL6, IL10 및 IFN(인터페론)-감마를 포함한 다양한 사이토카인에 의해 유도된다.
IL-6, Epo, GCSF 및 렙틴의 시그널링에서는 SOCS3의 각 사이토카인 수용체와의 결합이 SOCS3의 억제 기능에 매우 중요한 것으로 밝혀졌다.
SOCS3의 과잉발현은 지방조직과 간에서 인슐린 신호를 억제하지만 [10]근육에서는 억제하지 않는다.그러나 생쥐의 골격근에서 SOCS3가 삭제되면 비만과 관련된 인슐린 [10]저항성을 예방할 수 있다.
SOCS3는 세라마이드 [11]합성의 증가로 인해 렙틴 저항성과 인슐린 저항성에 모두 기여한다.이 때문에 SOCS 유전자를 제거하면 비만의[10] 인슐린 저항성을 막을 수 있다는 연구결과가 나왔다.
이 유전자의 생쥐에 대한 연구는 태아 간 조혈의 음성 조절과 태반 [12]발달에서 이 유전자의 역할을 제시했다.
SOCS3 단백질은 JAK2 키나제에 결합할 수 있으며, JAK2 키나제의 활성을 억제합니다.
